B. licheniformis의 설사원인균의 성장 억제 효과

B. licheniformis의 E. coli HGU-3 및 장내세균의 유해효소 억제효과

1. B. licheniformis / S. sonnei

E.coli HGU-3 가 생산하는 유해효소인 β-glucuronidase 효소활성은 E.coli HGU-3와 B. licheniformis 를 1:1로 혼합배양한 경우는 거의 활성의 변화가 없었으나 1:10인 경우는 효소활성이 29% 저해되었으며, Tryptophanase효소활성은 혼합배양한 경우 거의 변화가 없었다.

- 장내세균 유래 유해효소인 β-glucuronidase 효소활성은 B. licheniformis와 장내균총의 균수가 1:10³ 인 경우 45%의 저해활성을 보였으며, 1:10² 인 경우 23%의 저해활성을 보였다.
Tryptophanase 효소활성은 B. licheniformis의 농도변화에 따라 차이를 보이기도 하였으나 대부분의 경우 5~20% 정도의 저해활성을 보였다. Urease의 저해활성도 20% 정도의 저해활성을 보였다. 그 외의 β-glucuronidase의 경우는 50% 정도 저해하는 경향을 보였다.

B.licheniformis와 설사 원인균 동시 배양 시 유해균의 성장에 미치는 효과

B. licheniformis 와 E.coli O-157, S.typhi 또는 S.sonnei 를 1:10의 비율로 동시에 이식하여 배양하였다. 이식 배양 6시간부터 대부분의 설사원인 균주가성장하기 시작하나 12시간에서는 B. licheniformis 의 성장이 활발해지면서 설사원인 균주들의 성장이 크게 억제되었다. 배양 12시간과 24시간에서 S.typhi 는 대조군에 비해 약 90% 이상의 억제를 보였으며, S. sonnei 만을 이식한 군에 비해 95% 이상의 성장억제를 보였으며, E,coli O-157도 50% 이상의 성장억제를 보였다.

B. licheniformis 의 세포벽성분에서는 감염억제 효과가 거의 없었으나, 세포질 성분에서는 우수한 Rotarirus 감염 억제효과를 나타냈다.

A, Brain-stem tissue from healthy and bovine spongiform encephalopathy (BSE)-infected cow were pretreated at 115℃. Different aliquots after the heat pretreatment were differently processed: whole homogenates (lanes 1, 2, 5, 6, 9, 10, 13, and 14), supernatants collected from centrifugation at 2000g for 15min(lanes 3, 4, 7, 8, 11, 12, 15, and 16), and aliquots frozen for 17days(lanes 2, 4, 6, 8, 10, 12, 14, and 16).
B, The effect of preheating in the presence of N-lauroylsarcosine (sarc) on subsequent digestion with keratinase (KE) is shown. Samples were not precipitated with methanol.
C, the effect of detergents is shown. Before heating, homogenates from a BSE-infected animal were prepared in 2 different lysis buffers with different detergent compositions:2% N-lauroylsarcosine (S) vs. 0.5% Triton X-100 and 0.5% sodium deoxycholate (T). MATERIAL was studied both without and after digestion with KE. The tissue equivalents applied were 1000㎍, except 200㎍ in lane N of panel A and 400㎍ in all lanes of panel B. Arrows indicate the position of molecular-weight markers carbonic anhydrase (29 kDa) and β-lactoglobulin (18.4 kDa). All homogenates were prepared in the presence of 2% N-lauroylsarcosine, except where indicated. +, infected with BSE; -, healthy; N, sample containing nonheated nondigested brain-stem material from a healthy cow (control for intact PrP); P, unheated sample from a BSE-infected cow digested with proteinase K(PrPres control).
